I've just picked up and Australian cassette on the CBS label by Alison Moyet which has 5 12" mixes on it. The same tracks are on both sides, does this count as a single?

A mix or a medley or something alike on each side is mostly seen as a single in my opinion.
The running time is a little against it, but I will say yes.

Don't see why not - there are plenty of 12" singles which have five (or even more) remixes on them.
